Product Description

The rabbit-derived polyclonal anti-propionyl-mono-methyllyine antibodies are crosslinked to agarose with stable amide linkages. The antibody agarose conjugated beads is capable of capturing the anti-propionyl-mono-methyllyine peptides. These antibody beads were used in a novel chemical proteomics approach for global screening for mono-methyllysine substrates (1).

Scientific Description

The reversible lysine methylation of proteins plays a vital role in the regulation of many cellular processes including chromatin dynamics and gene transcription etc. The lysine residue can be modified at its ɛ-amino side-chain by mono, di-, and tri-methyl groups. The protein substrates of Kme remain largely unknown due to the relatively low energy radiation emitted by 3H or 14C makes it difficult to detect Kme substrates by a radio labeling approach. To overcome this technical challenge, we develop a chemical proteomics approach for global screening for kme substrates. We first chemically derivatized the mono-methyl ε-amine group of the lysine residue, adding a propionyl moiety. Then, the propionyl mono-methylated peptides were enriched using the pan anti-propionyl mono-methyllysine antibody conjugated agarose beads, followed by HPLC/MS/MS analysis. We have identified 446 Kme sites in 398 proteins, including 3 novel histone Kme marks.  


Storage & Stability

Store product at -20oC. Avoid repeated freeze/thaw. Antibody is supplied in PBS with 50% glycerol and 0.01% sodium azide.Stable for 12 months from date of receipt.
