Product Description

The pan polyclonal anti-2-hydroxyisobutyryllysine (anti-2ohibutyryllysine) antibody is produced by immunizing rabbits with 2ohibutyrylated BSA. Antibodies are purified by 2ohibutyryl-lysine agarose affinity chromatography. The product specifically recognizes proteins with lysine 2ohibutyrylated residues but not the structurally-similar modified lysine residues or non-modified lysine residues. Besides common applications in Western blotting, the antibody has been well utilized for proteomic screening to lysine 2ohibutyrylated substrates from protease-digested proteins both in prokaryotic and eukaryotic cells.  

 

Source/Purification

The pan monoclonal anti-2-hydroxyisobutyryllysine antibody is produced by immunizing mice with 2-hydroxyisobutyrylated BSA. Antibodies are purified by 2-hydroxyisobutyryllysine agarose affinity chromatography.

Recommended antibody dilution

WB: 1:1000

Important

For WB, incubate membrane with diluted antibody in 5% nonfat milk, 1 x TBS, 0.1% Tween-20 for two hours at room temperature with gentle shaking. Prepare working dilution immediately before use. Use at

an assay dependent concentration. Optimal dilutions/concentrations should be determined by the end user. Not yet tested in other applications.

Scientific Description


Histones are subjected to a variety of enzyme catalyzed modifications, including acetylation, methylation, ubiquitylation, etc. 2-hydroxyisobutyrylation of lysine is a newly identified reversible modification controlling chromosome structure and gene transcription. The reversible lysine 2-hydroxyisobutyrylation has been well demonstrated in eukaryotic histones from worm to human. The unique structure and genomic localization of histone lysine 2-hydroxyisobutyrylation suggest that it is mechanistically and functionally different from histone lysine acetylation.


 

Storage & Stability

The antibody is kept in PBS with 50% glycerol and 0.01% sodium azide. Upon receipt, please centrifuge the antibody at 12,000 x g for 20 seconds and store the antibody at -20°C. Avoid repeated freeze/thaw cycles. Stable for 12 months from date of receipt. Leave the antibody at room temperature for 2 minutes and gently mixed using pipette before usage.
