Product Description

The rabbit-derived pan anti-mono-, dimethyllysine antibody is purified with Protein A-conjugated agarose followed by methyl lysine agarose affinity chromatography. It specifically recognizes proteins with mono-, dimethyl lysine residues but not the trimethyl lysine, acetyl lysine or unmodified lysine residues. In addition to the common applications such as Western blotting, the antibody is capable of capturing lysine mono-, dimethylated peptides from protease-digested proteins both in prokaryotic and eukaryotic cells in systematically proteomic screening.

 

Source/Purification

The rabbit polyclonal antibody is produced by immunizing animals with methylated-KLH conjugates. Antibody is purified with Protein A-conjugated agarose followed by methyl lysine agarose affinity chromatography.

Recommended antibody dilution

WB: 1:1000; IAP: 1:200

Important

For Western blotting, incubate membrane with diluted antibody in 5% BSA, 1 x TBS, 0.1% Tween-20 at 4℃ with gentle shaking, overnight

Use at an assay dependent concentration. Optimal dilutions/concentrations should be determined by the end user. Not yet tested in other applications.

Anti-mono/di-methyllysine antibody

A. Dot blotting analysis on indicated amount of lysine methylated-KLH conjugates (lane 1), trimethylated-KLH conjugates (lane 2), acetylated-KLH conjugates (lane 3) and unmodified KLH (lane 4) using anti-mono-, dimethyllysine polyclonal antibody.


B. Western blotting analysis on 20 μg of crude proteins from HeLa whole cell lysates using anti-mono-, dimethyllysine polyclonal antibody (duplicated loading, 1:2000).


Scientific Description

The post-translational ε-amino lysine methylation of proteins, like acetylation and phosphorylation, is an important reversible modification controlling protein activity. The reversible lysine methylation of proteins plays a vital role in the regulation of many cellular processes including chromatin dynamics and gene transcription etc. Methylation of lysine residues is modulated by specific counteractive enzymes including lysine methylases (KMTs) and demethylases (KDMs). Mono-, dimethylation is a methylated form of lysine and occurs in both histones and non-histone substratres. Mono-, dimethylation in histones regulated by specific histone methylases (HMTs) and histone demethylases (HDMs) is termed as “histone code” or “epigenetic code”, whose pathway has become promising targets for discovery of anti-cancer drugs.


Storage & Stability

Store product at -20oC. Avoid repeated freeze/thaw. Antibody is supplied in PBS with 50% glycerol and 0.01% sodium azide.Stable for 12 months from date of receipt.
