Product Description

The rabbit-derived pan anti-trimethyllysine antibody is purified with Protein A-conjugated agarose followed by trimethyl lysine agarose affinity chromatography. It specifically recognizes proteins with trimethyl lysine residues but not the mono-, dimethyl lysine, acetyl lysine or unmodified lysine residues. In addition to the common applications such as Western blotting, the antibody is capable of capturing lysine trimethylated peptides from protease-digested proteins both in prokaryotic and eukaryotic cells in systematically proteomic screening.


Source/Purification

The rabbit polyclonal antibody is produced by immunizing animals with trimethylated-KLH conjugates. Antibody is purified with Protein A-conjugated agarose followed by trimethyl lysine agarose affinity chromatography.

Recommended antibody dilution

Important

For Western blotting, incubate  membrane with diluted antibody in 5% nonfat milk, 1 x TBS, 0.1% Tween-20 for  two hours at room temperature with gentle shaking.  

Use at an assay dependent concentration. Optimal  dilutions/concentrations should be determined by the end user.  

Anti-tri-methyllysine rabbit pAb

A. Dot blotting analysis on indicated amount of lysine trimethylated-KLH conjugates (lane 1), mono-, dimethylated-KLH conjugates (lane 2), acetylated-KLH conjugates (lane 3) and unmodified KLH (lane 4) using anti-trimethyllysine polyclonal antibody.


B. Western blotting analysis on 20 μg of crude proteins from HeLa whole cell lysates using anti-trimethyllysine polyclonal antibody (duplicated loading, 1:2000).


Scientific Description

The post-translational ε-amino lysine methylation of proteins, like acetylation and phosphorylation, is an important reversible modification controlling protein activity. The reversible lysine methylation of proteins plays a vital role in the regulation of many cellular processes including chromatin dynamics and gene transcription etc. Methylation of lysine residues is modulated by specific counteractive enzymes including lysine methylases (KMTs) and demethylases (KDMs). Trimethylation is a methylated form of lysine and occurs in both histones and non-histone substratres. Trimethylation in histones regulated by specific histone methylases (HMTs) and histone demethylases (HDMs) is termed as “histone code” or “epigenetic code”, whose pathway has become promising targets for discovery of anti-cancer drugs.


Storage & Stability

Store product at -20oC. Avoid repeated freeze/thaw. Antibody is supplied in PBS with 50% glycerol and 0.01% sodium azide.Stable for 12 months from date of receipt.
