Product Description

The rabbit-derived pan propionyl-mono-methyllysine antibody is purified with propionyl-mono-methylated-lysine agarose affinity chromatography. It specifically recognizes peptides with propionyl-mono-methyllysine residues but not the mono-methylllysine or other structural similar residues. This antibody is used in a novel chemical proteomics approach for global screening for mono-methyllysine substrates (1).  


Source/Purification

This antibody is produced by immunizing rabbits with propionyl- mono-methyllysine-conjugated keyhole limpet hemocyanin (KLH) and purified by propionyl-methylated-lysine agaroseaffinity chromatography.

A. Dot blotting analysis on propionyl-mono-methylated peptide library (Lane 1), mono-methylated peptide library (Lane 2), di-methylated peptide library (Lane 3), tri-methylated peptide library (Lane 4), propionylated peptide library (Lane 5), butyrylated peptide library (Lane 6), crotonylate peptide library (Lane 7), and unmodified peptide library (Lane 8) using anti-propionyl-mono-methyllysine rabbit pAb.

 

B. Mass spectrometer analysis: Representative annotated ion trap CID spectrum of a propionyl-13CD3-methyllysine-bearing peptide from protein HSP90. Crude proteins from HeLa cell werepropionylated, then digested by trypsin. The propionyl-mono-methyllysine remnant-containing peptides were affinity enriched by the anti-propionyl-mono-methyllysine rabbit pAb followed by LC-ESI-MS/MS in a LTQ Orbitrap Elite mass spectrometry. The sequence of the representative propionyl-mono-methylated peptide is indicated and the fragment ions are labeled in the MS/MS spectrum.  


Scientific Description

The reversible lysine methylation of proteins plays a vital role in the regulation of many cellular processes including chromatin dynamics and gene transcription etc.The lysine residue can be modified at its ɛ-amino side-chain by mono, di-, and tri-methyl groups. The protein substrates of Kme remain largely unknown due to the relatively low energy radiation emitted by 3H or 14C makes it difficult to detect Kme substrates by a radiolabeling approach.  s. To overcome this technical challenge,we develop a chemical proteomics approach for global screening for kme substrates. We first chemically derivatized the mono-methyl ε-amine group of the lysine residue, adding a propionyl moiety. Then, the propionyl mono-methylated peptides were enriched using the pan anti-propionyl mono-methyllysine antibody, followed by HPLC/MS/MS analysis.We have identified 446 Kme sites in 398 proteins, including 3 novel histone Kme marks.
