Product Description

The rabbit-derived pan butyryllysine antibody is purified with butyryl-lysine agarose affinity chromatography. It specifically recognizes proteins with butyryllysine residues but not the acetyllysine or propionyllysine residues with structural similarity. Except the common applications such as Western blotting, immunoprecipitation and immunofluorescence, the antibody is capable of capturing lysine butyrylated peptides from protease-digesting proteins both in prokaryotic and eukaryotic cells in systematically proteomic screening.  


Source/Purification

This product is produced by immunizing rabbits with butyrylated BSA. Antibodies are purified by butyrylated-lysine agarose affinity chromatography.

Important

For Western blotting, incubate  membrane with diluted antibody in 5% nonfat milk, 1 x TBS, 0.1% Tween-20 for  two hours at room temperature with gentle shaking.  

Use at an assay dependent concentration. Optimal  dilutions/concentrations should be determined by the end user.

Scientific Description

Butyrylation of lysine, structurally similar to lysine acetylation and lysine propionylation, is an newly identified reversible modification controlling protein activity. With integrated proteomic approaches and biochemistry analysis, lysine butyrylation has been well demonstrated in both prokaryotes and eukaryotes in wide ranges of proteins including histones and non-histone substrates. Given the facts that many lysine residues in histones and non-histone substrates, such as p53, p300/CBP, are butyrylated, lysine butyrylation may play a vital role in epigenetic modulation by impacting on chromatin dynamics and plasticity, DNA transcriptional regulation and tumorigenesis, etc.


Storage & Stability

Store product at -20oC. Avoid repeated freeze/thaw. Antibody is supplied in PBS with 50% glycerol and 0.01% sodium azide.Stable for 12 months from date of receipt.
