Product Description

The pan β-hydroxybutyryllysine rabbit monoclonal antibody was produced by immunizing rabbits with 3ohbutyrylated KLH conjugates. Antibodies are purified by protein A. It specifically recognizes β-hydroxybutyrylated proteins but not other structural similar modified proteins such as acetylated, crotonylated.

Important

For Western blotting, incubate  membrane with diluted antibody in 5% nonfat milk, 1 x TBS, 0.1% Tween-20 for  two hours at room temperature with gentle shaking.  

Use at an assay dependent concentration. Optimal  dilutions/concentrations should be determined by the end user.

Scientific Description

Histones are subjected to a variety of enzyme catalyzed modifications, including acetylation, methylation, phosphorylation, ubiquitylation, etc. β-hydroxybutyrylation of lysine is a newly identified reversible modification controlling chromosome structure and gene transcription. The reversible lysine β-hydroxybutyrylation has been well demonstrated in eukaryotic histones from worm to human. The unique structure and genomic localization of histone lysine β-hydroxybutyrylation suggest that it is mechanistically and functionally different from histonelysine acetylation. Specifically, in both human somatic and mouse male germ cell genomes, histone β-hydroxybutyrylation marks either active promoters or potential enhancers


Storage & Stability

Store product at -20 ℃. Avoid repeated freeze / thaw cycles. Storage Buffer: Antibody is kept in PBS with 50% glycerol and 0.01% sodium azide. Stable for 12 months from date of receipt.
