Product Description

Important

For Western blotting, incubate  membrane with diluted antibody in 5% nonfat milk, 1 x TBS, 0.1% Tween-20 for  two hours at room temperature with gentle shaking.  

Use at an assay dependent concentration. Optimal  dilutions/concentrations should be determined by the end user.  

Anti-β-hydroxybutyryllysine rabbit pAb

A. Dot blotting analysis on 10 ng of lysine 3ohbutyrylated peptide library (A1), lysine 3ohbutyrylated BSA (B1), selected histone lysine 3ohbutyrylated peptides (A2-5, B2-5, C1-5), unmodified lysine peptide library (A6), unmodified BSA (B6), lysine acetylated peptide library (C6), lysine propionylated peptide library (A7), lysine butyrylated peptide library (B7), lysine 2ohibutyrylated peptide library (C7), lysine crotonylated peptide library (A8) and randomly selected histone unmodified peptides (B8, C8) using anti-β-hydroxybutyryllysine rabbit pAb (1:1000).


B. Western blotting analysis on 30 μg of crude proteins from HeLa, C2C12, COS total cell lyates and HeLa cell with (right) or without (left) sodium 3ohbutyrylate treatment (100 mM,72 hr) using anti-β-hydroxybutyryllysine rabbit pAb.


Scientific Description

Histones are subjected to a variety of enzyme catalyzed modifications, including acetylation, methylation, phosphorylation, ubiquitylation, etc. β-hydroxybutyrylation of lysine is a newly identified reversible modification controlling chromosome structure and gene transcription. The reversible lysine β-hydroxybutyrylation has been well demonstrated in eukaryotic histones from worm to human. The unique structure and genomic localization of histone lysine β-hydroxybutyrylation suggest that it is mechanistically and functionally different from histonelysine acetylation. Specifically, in both human somatic and mouse male germ cell genomes, histone β-hydroxybutyrylation marks either active promoters or potential enhancers


Storage & Stability

Store product at -20 ℃. Avoid repeated freeze / thaw cycles. Storage Buffer: Antibody is kept in PBS with 50% glycerol and 0.01% sodium azide. Stable for 12 months from date of receipt.
