Product Description

This product is produced by immunizing rabbits with a synthetic mono-methyl peptide corresponding to residues surrounding Arg19 of human histone H4. Antibodies are purified by protein A-conjugated agarose followed by mono-methylated histone H4 (Arg19) peptide affinity chromatography.

Important

For Western blotting, incubate  membrane with diluted antibody in 5% nonfat milk, 1 x TBS, 0.1% Tween-20 for  two hours at room temperature with gentle shaking.  

Use at an assay dependent concentration. Optimal  dilutions/concentrations should be determined by the end user.

A.Dot blotting analysis on indicated amount of mono-methylated H4 peptide at Arg19 (lane 1) , mono-methylated H4 peptide at Arg3 (lane 2), symmetric di-methylated H4 peptide at Arg3 (lane 3), asymmetric di-methylated H4 peptide at Arg3 (lane 4), mono-methylated H4 peptide at Arg17 (lane 5), mono-methylated H4 peptide at Lys23 (lane 6), and unmodified H4 peptide at Arg19 (lane 7) using mono-methyl-histone H4 (Arg19) rabbit pAb.


B. Western blotting analysis on 30 μg of crude proteins from HeLa, H1299, 293T and C2C12 whole cell lysates using mono-methyl-histone H4 (Arg19) rabbit pAb (1:2000).


C. Western blotting analysis on 30 μg of crude proteins from mouse liver, mouse spleen tissue lysate using mono-methyl-histone H4 (Arg19) rabbit pAb (1:500).


D. Western blotting  analysis on 30 μg of crude proteins from Rice histone lysate, N2a and Spleen whole cell lysates using mono-methyl-histone H4 (Arg19) rabbit pAb (1:400).




Scientific Description

Proteins post-translational Modification (PTM) is an important reversible event controlling protein activity. The amino-terminal tails of core histones undergo multiple modifications in multiple sites, termed as “histone code” or “epigenetic code”. Lysine methylation or arginine methylation in core histones is a major determinant for the formation of active and inactive regions of the genome and therefore plays vital roles in multiple cellular events. In most species, arginine methylation occurs primarily on histones H3 (Arg2, 8, 17, 26) and H4 (Arg3) and has been implicated in both transcriptional activation and silencing. Arginine methylation in histones is carried out by the arginine N-methyltransferase (PRMT), which catalyzes the transfer of a methyl group from S-adenosylmethionine (AdoMet) to a guanidine nitrogen of arginine. Arginine methylation in histones is impaired in the pathologies of cancer and other diseases and therefore, enzymes regulating histone arginine methylation have become promising targets for anti-cancer drugs.


Storage & Stability

Store product at -20℃. Avoid repeated freeze/thaw. Antibody is supplied in PBS with 50% glycerol and 0.01% sodium azide. Stable for 12 months from date of receipt.
