Product Description
The mouse-derived antibody is purified with protein G-conjugated agarose followed by asymmetric-di-methylated histone H2A at arginine3 peptide affinity chromatography. As the amino acid sequence around Arg3 (SGRGK) for histone H4 and H2A is same, this antibody recognizes both histone H2A and H4 with asymmetric di-methylation at Arg3.  
Source/Purification
This antibody is produced by immunizing Balb/c mice with a synthetic di-methyl(asy) peptide corresponding to residues surrounding Arg3 of human histone H2A. Antibodies are purified by protein G-conjugated agarose followed by asymmetric di-methylated histone H2A at arg3 peptide affinity chromatography.

Important
For Western blotting, incubate membrane with diluted antibody in 5% nonfat milk, 1 x TBS, 0.1% Tween-20 for two hours at room temperature with gentle shaking.  

Use at an assay dependent concentration. Optimal dilutions/concentrations should be determined by the end user. Not yet tested in other applications.  

Anti-di-methyl(asy)-Histone H4 H2A (Arg3) mouse mAb

A. Dot blotting analysis on indicated amount of asymmetric di-methylated H2A peptide at Arg3 (lane 1), asymmetric di-methylated H4 peptide at Arg3 (lane 2), mono-methylated H2A peptide at Arg3 (lane 3), acetylated H2A peptide at Arg5 (lane 4), butyrylated H2A peptide at Arg5 (lane 5), 2ohibutyrylated H2A peptide at Arg5 (lane 6), 3-ohbutyrylated H2A peptide at Lys5 (lane 7) and unmodified H2A peptide at Arg3 (lane 8) using di-methyl(asy)-histone H2A/H4 (Arg3) mouse mAb.  
B. Western blotting analysis on 30 μg of crude proteins from K562, MCF-7, C2C12, COS-7 whole cell lysates using asymmetric di-methyl-histone H2A/H4 (Arg3) mouse mAb (1:2000).   
Scientific Description
Proteins post-translational Modification (PTM) is an important reversible event controlling protein activity. The amino-terminal tails of core histones undergo multiple modifications in multiple sites, termed as “histone code” or “epigenetic code”. Lysine methylation or arginine methylation in core histones is a major determinant for the formation of active and inactive regions of the genome and therefore plays vital roles in multiple cellular events. In most species, arginine methylation occurs primarily on histones H3 (Arg2, 8, 17, 26) and H4 (Arg3) and has been implicated in both transcriptional activation and silencing. Arginine methylation in histones is carried out by the arginine N-methyltransferase (PRMT), which catalyzes the transfer of a methyl group from S-adenosylmethionine (AdoMet) to a guanidine nitrogen of arginine. Arginine methylation in histones is impaired in the pathologies of cancer and other diseases and therefore, enzymes regulating histone arginine methylation have become promising targets for anti-cancer drugs.  
Storage & Stability
The antibody is delivered in powder form. After dissolved with the antibody reconstitution buffer, please store the antibody at -20°C. Avoid repeated freeze/thaw cycles. Stable for 12 months from date of receipt.
