Product Description

The mouse-derived antibody is purified with protein G-conjugated agarose. It specifically recognizes histone H3 only when it tri-methylated at Lys9 and phosphorylated at Ser10.  


Source/Purification

This product is produced by immunizing mice with a synthetic tri-metylated and phosporylated peptide corresponding to residues surrounding Lys9 and ser10 of human histone H3. Antibodies are purified by protein G-conjugated agarose.

Important

For Western blotting, incubate membrane with diluted antibody in 5% nonfat milk, 1 x TBS, 0.1% Tween-20 for two hours at room temperature with gentle shaking.  

Use at an assay dependent concentration. Optimal dilutions/concentrations should be determined by the end user.

A. Dot blotting analysis on indicated amount of tri-methyl(Lys9)/phospho(Ser10) H3 peptide (lane 1), phosphorylated H3 peptide at Thr3 (lane 2), crotonylated H3 peptide at Lys4 (lane 3), mono-methylated H3 peptide at Lys4 (lane 4), di-methylated H3 peptide at Lys4 (lane 5), tri-methylated H3 peptide at Lys4 (lane 6), phosphorylated H3 peptide at Thr6 (lane 7), acetylated H3 peptide at Lys9 (lane 8), butyrylated H3 peptide at Lys9 (lane 9), 2ohibutyrylated H3 peptide at Lys9 (lane 10), 3ohbutyrylated H3 peptide at Lys9 (lane 11), mono-methylated H3 peptide at Lys9 (lane 12), di-methylated H3 peptide at Lys9 (lane 13), tri-methylated H3 peptide at Lys9 (lane 14), phosphorylated H3 peptide at Ser10 (lane 15), crotonylated H3 peptide at Lys14 (lane 16), mono-methylated H3 peptide at Lys14 (lane 17), di-methylated H3 peptide at Lys14 (lane 18), and unmodified H3 peptide at Lys9/Ser10 (lane 19) using anti-tri-methyl (Lys9)/ phosphor(ser10) histone H3 mouse mAb.  


B. Western blotting analysis on 30 μg of crude proteins from human HeLa whole cell lysates with (Lane 2) or without (Lane 1) treatment of nocodazole (100 ng/ml, 18hr) and calyculin A ( CA, 100 nM, 1hr) using anti-tri-methyl (Lys9)/ phosphor(ser10) histone H3 mouse mAb.  


Scientific Description

Histones are subjected to a variety of enzyme catalyzed modifications, including acetylation, methylation, ubiquitylation and newly discovered succinylation, crotonylation and 2-hydroxyisobutyrylation, etc. These modifications have a direct effect on the accessibility of chromatin to transcription factors and, therefore, on gene expression. Histone H3 is found to be phosphorylated during both mitosis and meiosis.  It is believed that H3 phosphorylation is involved in transcriptional activation during cell division. Histone H3 phosphorylation may be initiated at different phases of the cell division in different organisms, and Histone H3 of metaphase chromosomes are always found to be heavily phosphorylated.  


Storage & Stability

 The antibody is delivered by powder. After dissolved with the antibody constitution buffer, please store it at -20oC.
