Product Description

The mouse-derived antibody is purified with protein G-conjugated agarose followed by butyrylated histone H3 (Lys9) peptide affinity chromatography. It specifically recognizes histone H3 with butyrylation at Lys9.  



Source/Purification

This product is produced by immunizing mice with a synthetic butyryl peptide corresponding to residues surrounding Lys9 of human histone H3. Antibodies are purified by protein G-conjugated agarose followed by butyrylated histone H3 (Lys9) peptide affinity chromatography.

Important

For Western blotting, incubate  membrane with diluted antibody in 5% nonfat milk, 1 x TBS, 0.1% Tween-20 for  two hours at room temperature with gentle shaking.  

Use at an assay dependent concentration. Optimal  dilutions/concentrations should be determined by the end user.

A. Dot  blotting analysis on indicated amount of butyrylated H3 peptide at Lys9 (lane  1), crotonylated H3 peptide at Lys4 (lane 2), acetylated H3 peptide at Lys9  (lane3), crotonylated H3 peptide at Lys9 (lane 4) and unmodified H3 peptide  (lane 5) using butyryl-histone H3 (Lys9) mouse mAb.


B. Western blotting analysis on 30 μg of crude proteins  from HeLa whole cell lysates with (right) or without (left) sodium butyrate  treatment (30 mM, 4 hours) using butyryl-histone H3 (Lys9) mouse mAb (1:2000).   


Scientific Description

Histones are subject to a variety of enzyme catalyzed modifications, including acetylation, methylation, phosphorylation, ubiquitylation, etc. Butyrylation of lysine, structurally similar to lysine acetylation, is a newly identified reversible modification controlling protein activity. The reversible lysine butyrylation has been well demonstrated in both prokaryotes and eukaryotes in wide ranges of proteins including histones and non-histone substrates, such as p53. Similar to acetylation of histione H4 at lysine 12, butyrylation of histone H3 at lysine 9 may play a vital role in the epigenetic modulation, including chromatin remodeling, and DNA transcriptional regulation.  



Storage & Stability

Store product at -20oC. Avoid repeated freeze/thaw. Antibody is supplied in PBS with 50% glycerol and 0.01% sodium azide.Stable for 12 months from date of receipt.
