Product Description

The mouse-derived antibody is purified with protein G-conjugated agarose followed by butyrylated Histone H3 at Lys27 peptide affinity chromatography. It specifically recognizes Histone H3 with butyrylation at Lys27.

 

Source/Purification

This product is produced by immunizing mice with a synthetic butyryl peptide corresponding to residues surrounding Lys27 of human histone H3. Antibodies are purified by protein G-conjugated agarose followed by butyrylated histone H3 (Lys27) peptide affinity chromatography.

Important

For Western blotting, incubate membrane with diluted antibody in 5% nonfat milk, 1 x TBS, 0.1% Tween-20 for two hours at room temperature with gentle shaking.  

Use at an assay dependent concentration. Optimal dilutions/concentrations should be determined by the end user.

A. Dot blotting analysis on indicated amount of butyrylated H3 peptide at Lys27 (lane 1), acetylated H3 peptide at Lys23 (lane 2), propionylated H3 peptide at Lys23 (lane 3), butyrylated H3 peptide at Lys23 (lane 4), crotonylated H3 peptide at Lys23 (lane 5), acetylated H3 peptide at Lys27 (lane 6), crotonylated H3 peptide at Lys27 (lane 7), 2ohibutyrylated H3 peptide at Lys27 (lane 8), 3ohbutyrylated H3 peptide at Lys27 (lane 9), acetylated H3 peptide at Lys36 (lane 10), crotonylated H3 peptide at Lys36 (lane 11), and unmodified H3 peptide at Lys27 (lane 12) using butyryl-Histone H3 (Lys27) mouse mAb.  

 

B. Western blotting analysis on 30 μg of crude proteins from HeLa whole cell lysates with (right) or without (left) treatment of sodium butyrate (30 mM, 4 hours) using butyryl-Histone H3 (Lys27) mouse mAb (1:2000).  

 

Scientific Description

Histones are subjected to a variety of enzyme catalyzed modifications, including acetylation, methylation, phosphorylation, ubiquitylation, etc. Butyrylation of lysine is a newly identified reversible modification controlling chromosome structure and gene transcription. Lysine butyrylation is highly conserved in eukaryotic cells from worm to human. The unique structure and genomic localization of histone lysine butyrylation suggest that it is mechanistically and functionally different from histone lysine acetylation. Specifically, in both human somatic and rabbit male germ cell genomes, histone butyrylation marks either active promoters or potential enhancers. Butyrylation of histone H3 at Lys27 may play a vital role in the epigenetic modulation, including chromatin remodeling and DNA transcriptional regulation.  


Storage & Stability

The antibody is delivered in powder form. After dissolved with the antibody reconstitution buffer, please store the antibody at -20°C. Avoid repeated freeze/thaw cycles. Stable for 12 months from date of receipt.
