Product Description

The rabbit-derived antibody is purified with protein A-conjugated agarose followed by crotonylated histone H3 (Lys9) peptide affinity chromatography. It specifically recognizes histone H3 with lysine crotonylation at Lys9.

 

Source/Purification

This product is produced by immunizing rabbits with a synthetic crotonyl peptide corresponding to residues surrounding Lys9 of human histone H3. Antibodies are purified by protein A-conjugated agarose followed by crotonylated histone H3 (Lys9) peptide affinity chromatography.

Important

For Western blotting, incubate  membrane with diluted antibody in 5% nonfat milk, 1 x TBS, 0.1% Tween-20 for  two hours at room temperature with gentle shaking.  

Use at an assay dependent concentration. Optimal  dilutions/concentrations should be determined by the end user.

A. Dot  blotting analysis on indicated amount of crotonylated H3 peptide at Lys9 (lane  1), butyrylated H3 peptide at Lys9 (lane 2), acetylated H3 peptide at Lys9  (lane 3), acetylated H3 peptide at Lys4 (lane4), crotonylated H3 peptide at  Lys4 (lane5) and unmodified H3 peptide at Lys9  (lane 6) using crotonyl-histone H3 (Lys9) rabbit pAb. 


B. Western blotting analysis on 30 μg of crude proteins from HeLa,  C2C12 and COS whole cell lysates using crotonylate-histone H3 (Lys9) rabbit pAb  (1:2000).


Scientific Description

Histones are subjected to a variety of enzyme catalyzed modifications, including acetylation, methylation, phosphorylation, ubiquitylation, etc. Crotonylation of lysine is a newly identified reversible modification controlling chromosome structure and gene transcription. The reversible lysine crotonylation has been well demonstrated in eukaryotic histones from worm to human. The unique structure and genomic localization of histone lysine crotonylation suggest that it is mechanistically and functionally different from histone lysine acetylation. Specifically, in both human somatic and mouse male germ cell genomes, histone crotonylation marks either active promoters or potential enhancers. Crotonylation of histone H3 at Lys12 may play a vital role in the epigenetic modulation, including chromatin remodeling and DNA transcriptional regulation.  


Storage & Stability

Store product at -20oC. Avoid repeated freeze/thaw. Antibody is supplied in PBS with 50% glycerol and 0.01% sodium azide.Stable for 12 months from date of receipt.
