Product Description

The mouse-derived antibody is purified with protein G-conjugated agarose followed by crotonylated histone H2B (Lys12) peptide affinity chromatography. It specifically recognizes histone H2B with crotonylation at Lys12.  


Source/Purification

This product is produced by immunizing mice with a synthetic crotonyl peptide corresponding to residues surrounding Lys12 of human histone H2B. Antibodies are purified by protein G-conjugated agarose followed by crotonylated histone H2B (Lys12) peptide affinity chromatography.

Important

For Western blotting, incubate  membrane with diluted antibody in 5% nonfat milk, 1 x TBS, 0.1% Tween-20 for  two hours at room temperature with gentle shaking.  

Use at an assay dependent concentration. Optimal  dilutions/concentrations should be determined by the end user.

A. Dot  blotting analysis on indicated amount of crotonylated H2B peptide at Lys12  (lane 1), butyrylated H2B peptide at Lys 5 (lane 2), crotonylated H2B peptide  at Lys11 (lane 3), acetylated peptide at Lys15 (lane 4), crotonylated H2B  peptide at Lys16 (lane 5), and unmodified H2B peptide at Lys12 (lane 6) using crotonyl-histone  H2B (Lys12) mouse mAb.


B. Western blotting analysis on 30 μg of crude proteins  from HeLa, C2C12 and COS whole cell lysates using crotonyl-histone H2B (Lys12) mouse  mAb (1:2000).  

 

Scientific Description

Histones are subjected to a variety of enzyme catalyzed modifications, including acetylation, methylation, phosphorylation, ubiquitylation, etc. Crotonylation of lysine is a newly identified reversible modification controlling chromosome structure and gene transcription. Lysine crotonylation is highly conserved in eukaryotic cellsfrom worm to human. The unique structure and genomic localization of histone lysine crotonylation suggest that it is mechanistically and functionally different from histone lysine acetylation. Specifically, in both human somatic and rabbit male germ cell genomes, histone crotonylation marks either active promoters or potential enhancers. Crotonylation of histone H2B at Lys12 may play a vital role in the epigenetic modulation, including chromatin remodeling and DNA transcriptional regulation.  


Storage & Stability

Store product at -20oC. Avoid repeated freeze/thaw cycles. Antibody is supplied in PBS with 50% glycerol and 0.01% sodium azide. Stable for 12 months from date of receipt.
