Product Description

The rabbit-derived antibody is purified with protein A-conjugated agarose followed by 2-hydroxyisobutyryl (2ohibutyryl) histone H2A (Lys5) peptide affinity chromatography. It specifically recognizes histone H2A with 2ohibutyrylation at Lys5.

 

Source/Purification

This product is produced by immunizing rabbits with a synthetic 2ohibutyryl peptide corresponding to residues surrounding Lys5 of human histone H2A. Antibodies are purified by protein A-conjugated agarose followed by 2ohibutyrylated histone H2A (Lys5) peptide affinity chromatography.

Important

For Western blotting, incubate membrane with diluted antibody in 5% nonfat milk, 1 x TBS, 0.1% Tween-20 for two hours at room temperature with gentle shaking.  

Use at an assay dependent concentration. Optimal dilutions/concentrations should be determined by the end user.

A. Dot blotting analysis on indicated amount of 2ohibutyrylated H2A peptide at Lys5 (lane 1), acetylated H2A peptide at Lys5 (lane 2), butyrylated H2A peptide at Lys5 (lane 3), 3ohbutyrylated H2A peptide at Lys5 (lane 4), acetylated H2A peptide at Lys9 (lane 5), succinylated H2A peptide at Lys9 (lane 6), 2ohibutyrylated H2A peptide at Lys9 (lane 7), 3ohbutyrylated H2A peptide at Lys9 (lane 8), and unmodified H2A peptide at Lys5 (lane 9) using 2ohibutyryl-histone H2A (Lys5) rabbit pAb.   


B. Western blotting analysis on 30 μg of crude proteins from HeLa whole cell lysates with (right) or without (left) treatment of sodium butyrate (30 mM, 4 hours) using 2ohibutyryl-histone H2A (Lys5) rabbit pAb (1:2000).  


C. Western blotting analysis on 30 μg of crude proteins from MCF-7 whole cell lysates with (right) or without (left) treatment of sodium butyrate (30 mM, 4 hours) using 2ohibutyryl-histone H2A (Lys5) rabbit pAb (1:2000).

 

Scientific Description

Histones are subjected to a variety of enzyme catalyzed modifications, including acetylation, methylation, phosphorylation, etc. 2-hydroxyisobutyrylation of lysine is a newly identified reversible modification regulating gene expression. The reversible lysine 2-hydroxyisobutyrylation has been well demonstrated in eukaryotic histones from worm to human. The unique structure and genomic localization of histone lysine 2-hydroxyisobutyrylation suggest that it is mechanistically and functionally different from histone lysine acetylation. Specifically, in both human somatic and mouse male germ cell genomes, histone2-hydroxyisobutyrylation marks either active promoters or potential enhancers.  

 

Storage & Stability

The antibody is delivered in powder form. After dissolved with the antibody reconstitution buffer, please store the antibody at -20°C. Avoid repeated freeze/thaw cycles. Stable for 12 months from date of receipt.
