Product Description

The mouse-derived antibody is purified with protein G-conjugated agarose followed by 2-hydroxyisobutyryl (2ohibutyryl) histone H3 (Lys14) peptide affinity chromatography. It specifically recognizes histone H3 with 2-ohibutyrylation at Lys14.



Source/Purification

This product is produced by immunizing mice with a synthetic 2-ohibutyryl peptide corresponding to residues surrounding Lys14 of human histone H3. Antibodies are purified by protein G-conjugated agarose followed by 2ohibutyrylated histone H3 (Lys14) peptide affinity chromatography.

Important

For Western blotting, incubate  membrane with diluted antibody in 5% nonfat milk, 1 x TBS, 0.1% Tween-20 for  two hours at room temperature with gentle shaking.  

Use at an assay dependent concentration. Optimal  dilutions/concentrations should be determined by the end user.

A. Dot blotting analysis on indicated amount of 2ohibutyrylated H3 peptide at Lys14 (lane 1), acetylated H3 peptide at Lys14 (lane 2), propionylated H3 peptide at Lys14 (lane 3), succinylated H3 peptide at Lys14 (lane 4), 3ohbutyrylated H3 peptide at Lys14 (lane 5), 2ohibutylated H3 peptide at Lys18 (lane 7), 3ohbutyrylated H3 peptide at Lys18 (lane 7) and unmodified H3 peptide at Lys14 (lane 8) using 2ohibutyryl-histone H3 (Lys14) mouse mAb.


B. Western blotting analysis on 30 μg of crude proteins from HeLa whole cell lysates with (lane 2) or without (lane 1) sodium butyrate (30 mM, 4hr) or sodium pyruvate (100 mM, 72hr) using 2ohibutyryl-histone H3 (Lys14) mouse mAb(1:2000).


Scientific Description

Histones are subjected to a variety of enzyme catalyzed modifications, including acetylation, methylation, phosphorylation, ubiquitylation, etc. 2-hydroxyisobutyrylation of lysine is a newly identified reversible modification controlling chromosome structure and gene transcription. The reversible lysine 2-hydroxyisobutyrylation has been well demonstrated in eukaryotic histones from worm to human. The unique structure and genomic localization of histone lysine 2-hydroxyisobutyrylation suggest that it is mechanistically and functionally different from histone lysine acetylation. Specifically, in both human somatic and mouse male germ cell genomes, histone 2-hydroxyisobutyrylation marks either active promoters or potential enhancers.



Storage & Stability

Store product at -20oC. Avoid repeated freeze/thaw. Antibody is supplied in PBS with 50% glycerol and 0.01% sodium azide.Stable for 12 months from date of receipt.
