Product Description

The rabbit-derived antibody is purified with protein A-conjugated agarose followed by 2-hydroxyisobutyryl (2-ohibutyryl) histone H3 (Lys79) peptide affinity chromatography. It specifically recognizes histone H3 with 2-ohibutyrylation at Lys79.   

Source/Purification

This product is produced by immunizing rabbits with a synthetic 2-ohibutyryl peptide corresponding to residues surrounding Lys79 of human histone H3. Antibodies are purified by protein A-conjugated agarose followed by 2ohibutyrylated histone H3 (Lys79) peptide affinity chromatography.

Important

For Western blotting, incubate membrane with diluted antibody in 5% nonfat milk, 1 x TBS, 0.1% Tween-20 for two hours at room temperature with gentle shaking.  

Use at an assay dependent concentration. Optimal dilutions/concentrations should be determined by the end user.

A. Dot  blotting analysis on indicated amount of 2-ohibutyryl H3 peptide at Lys79 (lane  1), succinylated H3 peptide at Lys79 (lane 2), 3-ohbutyrylated H3 peptide at Lys79  (lane 3) and unmodified H3 peptide at Lys79  (lane 4) using 2ohibutyryl-histone H3 (Lys79) rabbit pAb.  


B. Western blotting analysis on 30 μg of crude proteins  from HeLa whole cell lysates with (lane 2) or without (lane 1) sodium pyruvate  (100 mM, 72hr) using 2ohibutyryl-histone H3 (Lys79) rabbit pAb (1:2000).  

Scientific Description

Histones are subjected to a variety of enzyme catalyzed modifications, including acetylation, methylation, phosphorylation, ubiquitylation, etc. 2-hydroxyisobutyrylation of lysine is a newly identified reversible modification controlling chromosome structure and gene transcription. The reversible lysine 2-hydroxyisobutyrylation has been well demonstrated in eukaryotic histones from worm to human. The unique structure and genomic localization of histone lysine 2-hydroxyisobutyrylation suggest that it is mechanistically and functionally different from histone lysine acetylation. Specifically, in both human somatic and mouse male germ cell genomes, histone 2-hydroxyisobutyrylation marks either active promoters or potential enhancers.
