Product Description

The rabbit-derived antibody is purified with protein A-conjugated agarose followed by 2-hydroxyisobutyryl (2ohibutyryl) histone H4 (Lys20) peptide affinity chromatography. It specifically recognizes histone H4 with 2ohibutyrylation at Lys20.  


Source/Purification

This product is produced by immunizing rabbits with a synthetic 2ohibutyryl peptide corresponding to residues surrounding Lys20 of human histone H4. Antibodies are purified by protein A-conjugated agarose followed by 2ohibutyrylated histone H4 (Lys20) peptide affinity chromatography.

Important

 For Western blotting, incubate membrane with diluted antibody in 5% nonfat milk, 1 x TBS, 0.1% Tween-20 for two hours at room temperature with gentle shaking.  

 Use at an assay dependent concentration. Optimal dilutions/concentrations should be determined by the end user.

B.Western blotting analysis on 30 µg of crude proteins from HeLa whole cell lysates treatment with sodium sodium pyruvate (100 mM, 72hr) using 2ohibutyryl-histone H4 (Lys20) rabbit pAb (1:2000). Antibody incubated with 1.5 μM of the 2ohibutyrylated H4 at Lys20 peptides (lane 1) , antibody incubated withunmodified H4 peptides at Lys 20 (lane2), and no peptide blocking (lane 3).  


Scientific Description

Histones are subjected to a variety of enzyme catalyzed modifications, including acetylation, methylation, phosphorylation, etc. 2-hydroxyisobutyrylation of lysine is a newly identified reversible modification controlling chromosome structure and gene transcription. The reversible lysine 2-hydroxyisobutyrylation has been well demonstrated in eukaryotic histones from worm to human. The unique structure and genomic localization of histone lysine 2-hydroxyisobutyrylation suggest that it is mechanistically and functionally different from histone lysine acetylation. Specifically, in both human somatic and mouse male germ cell genomes, histone2-hydroxyisobutyrylation marks either active promoters or potential enhancers.  


Storage & Stability

The antibody is delivered in powder form. After dissolved with the antibody reconstitution buffer, please store the antibody at -20°C. Avoid repeated freeze/thaw cycles. Stable for 12 months from date of receipt.
