Product Description

The mouse-derived antibody is purified with protein G-conjugated agarose followed by β-hydroxybutyryl (3-ohbutyryl) histone H4 (Lys16) peptide affinity chromatography. It specifically recognizes histone H4 with β-hydroxybutyrylation at Lys16.


Source/Purification

This product is produced by immunizing mice with a synthetic 3-ohbutyryl peptide corresponding to residues surrounding Lys16 of human histone H4. Antibodies are purified by protein G-conjugated agarose followed by 3ohbutyryl histone H4 (Lys16) peptide affinity chromatography.

Recommended antibody dilution

Important

For Western blotting, incubate  membrane with diluted antibody in 5% nonfat milk, 1 x TBS, 0.1% Tween-20 for  two hours at room temperature with gentle shaking.  

Use at an assay dependent concentration. Optimal  dilutions/concentrations should be determined by the end user.

A. Dot blotting analysis on indicated amount of 3ohbutyrylated H4 peptide at Lys16 (lane 1), 3ohbutyrylated H4 peptide at Lys12 (lane 2), acetylated H4 peptide at Lys16 (lane 3), propionylated H4 peptide at Lys16 (lane 4), butyrylated H4 peptide at Lys16 (lane 5), 2ohibutyrylated H4 peptide at Lys16 (lane 6), 3ohbutyrylated H4 peptide at Lys20 (lane 7) and unmodified H4 peptide at Lys16 (lane 8) using β-hydroxybutyryl-histone H4 (Lys16) mouse mAb.  


B. Western blotting analysis on 30 μg of crude proteins from HeLa whole cell lysates with (lane 2) or without (lane 1) sodium 3-hydroxybutyrate (50 mM, 72hr) using β-hydroxybutyryl-histone H4 (Lys16) mouse mAb (1:2000).  


Scientific Description

Histones are subjected to a variety of enzyme catalyzed modifications, including acetylation, methylation, phosphorylation, ubiquitylation, etc. β-hydroxybutyrylation of lysine (Kbhb) is a newly identified histone marker that enriched in active gene promoters. Histone Kbhb marks are dramatically induced in response to elevated β-hydroxybutyrate levels in cultured cells and in livers from mice subjected to prolonged fasting or streptozotocin-induced diabetic ketoacidosis. Histone β-hydroxybutyrylation represents a new epigenetic regulatory mark that couples metabolism to gene expression, offering a new avenue to study chromatin regulation and diverse functions of β-hydroxybutyrate in the context of important human pathophysiological states, including diabetes, epilepsy, and neoplasia.  


Storage & Stability

Store product at -20 ℃. Avoid repeated freeze/thaw. Antibody is supplied in PBS with 50% glycerol and 0.01% sodium azide.Stable for 12 months from date of receipt.
